Abstract

The present study aimed at investigating and comparing the therapeutic effect of mesenchymal stem cells (MSCs), combined MSCs with ascorbic acid and SERCA1a gene transfected MSCs in STZ induced skeletal myopathy of male albino rat. Thirty five adult male albino rats were divided into: group Ї (Control Group) of 4 rats, group II (Diabetic Group) of 7 rats given single IP injection of STZ 50 mg/kg, group III (Mesenchymal Stem Cells Group) of 8 rats.7 rats were given STZ injection and 4 wks later 0.5 ml of cultured and labeled BMMSCs on two successive days. group IV (Mesenchymal Stem Cells and Ascorbic Acid Group) included 8 rats, 7 rats were given STZ and BMMSCs together with daily oral 500mg/Kg of AA. group V (SERCA1a Transfected Mesenchymal Stem Cells) of 8 rats, 7 rats were given STZ and 0.5 ml of cultured, labeled and SERCA1a lipofectamine transfected BMMSCs.Control and animals of groups II, III, IV and V were sacrificed 8 weeks from the start of the experiment. Gastrocnemius specimens were subjected to histological, immunohistochemical, morphometric ,biochemical, serological and statistical studies. GroupII revealed atypical widely separated fibers, disrupted myofibrils, few central nuclei, separated CT fibers, loss of striations of sarcoplasm, pale central nuclei, disorganized fibers and dark nuclei. Atypical fibers, markedly congested vessels,dense cellular infiltration and multiple macrophages were evident in the CT. group III showed some infiltrating cells, organized muscle fibers, peripheral nuclei, some central nuclei and few central flat dark nuclei, less separated fibers, obvious striations in some parts of the sarcoplasm, focal disorganization and focal disruption. Some atypical disrupted fibers were seen. group IV showed some fibers with rows of central nuclei, multiple central flat dark nuclei, some mononuclear cells, fibers with minimal separation of myofibrils, striations were seen in multiple areas of the sarcoplasm and focal disruption of myofibrils of some atypical fibers
