Abstract

The study was carried out in Ornamental Horticulture department, Faculty of agriculture, Cairo university, Giza, Egypt and tissue culture laboratories of ornamental plants and woody trees and biotechnology departments, national research centre (NRC), Dokki, Giza, Egypt during two successive seasons 2014 / 2015 and 2015/2016 to investigate the effect of some cytokines as (BA, Kin, 2ip), irradiations as physical mutagens and chemical mutagens on in vitro propagation behaviors morphological characters, as well as flowering of acclimatized plants, chemical constituents and anatomical structure of eustoma grandiflorum plant. The results showed that both number of shootlets formed per explant and number of leaves per shootlet were in highest values when BA was added to the culture medium at 0.4mg/l as compared to control. The longest shoots were obtained with MS culture medium supplemented with Kin (0.4mg/l). Gamma radiation: The experimental study was carried out on eustoma grandiflorum plant to evaluate the effect of gamma radiation as physical mutagenesis at different doses (0, 2, 5, 10, 20, 40, 60, 80, 100 and 120KR) using Co60 gamma source for 30min. on various morphological, flowering and anatomical characters.Using gamma irradiation at dose of 10 and 20KR gave the best results for shooting and rooting behaviors,highest survival percent of acclimatized plants, highest value of leaf area, highest rooting percentage and number of roots, increasing in chl. (a), chl.(a+b) and carotenoids content,while the dose of 2KR gave the highest value of chl.b content. Using the dose of 120KR recorded the best floral characters
