Abstract

Background and aim: The predominance of NAFLD is predicted to be in the near future as the principal cause of liver disease worldwide. The mechanism involves a complex cascade of events. As macroautophagy is a central mechanism for the breakdown of lipid droplet-stored TGs and cholesterol through a process termed lipophagy so modulation of autophagy may be of interest in treatment of NAFLD. Objectives: The present study was designed to investigate the potential role of CBZ as an autophagy modulator in NAFLD induced by High fat diet in rats. Materials and methods: Male wistar strain rats weighing (150-200g) were used. NAFLD induced by high fat diet (71% of energy from fat, 11% from carbohydrates, 18% from proteins) for twelve weeks. CBZ in a dose 25mg/kg and 100mg/kg/day were administrated orally daily for 4 weeks starting from the eighth week till the end of the experiment. the following parameters were measured in serum: FBG, insulin, ALT, AST, TG and cholesterol. Also, TG, cholesterol, SOD, AMPK and calcium were measured in hepatic tissue. Also, body weight and BP were measured. Histo-pathological examination of liver was performed in all groups. Results: Normal rats received CBZ didn{u2019}t produce any remarkable effects regarding all parameters measured except for liver weight ratio, liver enzyme (AST) which were significantly elevated in normal rats received both doses of drugs and serum cholesterol which were significantly elevated in normal rats received large dose
