Abstract

Background: Hepatitis C virus is a viral pandemic and a leading cause of chronic liver disease, an estimated 130-170 million people have HCV infection worldwide, prevalence is highest in Egypt with more than 10% of the general population are infected. The progression of hepatic disease in chronic viral hepatitis is accompanied by an increased production of reactive oxygen species (ROS), as well as an accumulation of oxidative DNA damage, which is primarily repaired through base excision repair. XRCC1 (X-ray repair cross complementing protein 1) is one of the most important proteins involved in this repair pathway. The study was carried out to verify the possible association of the XRCC1 rs25487 polymorphism with cirrhosis in chronic HCV patients. Subjects and Methods: A total 120 individuals were divided into 80 chronic HCV+ve patients (40 HCV +ve cirrhotic patients and 40 HCV +ve non cirrhotic patients) and 40 age and sex matched healthy control subjects were genotyped for XRCC1 gene (rs25487) polymorphism using Real-time PCR. Results: On comparing the two patients groups (cirrhotic and non cirrhotic) the results showed that there were no statistically significant difference in the distribution of different genotypes and alleles in XRCC1 A > G (GG and AG). Conclusion: XRCC1 polymorphism is not associated with increased susceptibility to cirrhosis in chronic HCV +ve patients
