Abstract

Diabetic encephalopathy is an important complication of diabetes characterized by cognitive impairment, neurochemical and structural abnormalities. This study aimed to evaluate the effect of coenzyme Q10 and niacin in comparison to reference drugs, against diabetic encephalopathy in rats. Diabetes mellitus was induced by a single intraperitoneal injection of streptozotocin (STZ 60 mg/kg). One month diabetic rats were treated with either coenzyme Q10 (10 mg/kg b.wt), niacin (40 mg/kg b.wt) and their combination, or glibenclamide (5 mg/kg b.wt), donepezil-hydrochloride (3 mg/kg b.wt) and their combination, for two consecutive weeks after diabetes induction. An elevation in blood glucose level (BGL) and a decrease in insulin level were evidenced in diabetic rats. Moreover, diabetes triggered an increment of malondialdehide (MDA) level along with a reduction of reduced glutathione (GSH) level and superoxide dismutase (SOD) activity. Increased levels of angiotensin II (Ang II), vascular cell adhesion molecule-1 (VCAM-1), intercellular adhesion molecule-1 (ICAM-1), interleukin-6 (IL-6), tumor necrosis factor alpha (TNF-Ü) and caspase-3 activity were observed in diabetic rats. Additionally, diabetic rats exhibited an elevation in acetylcholinesterase (AchE) activity associated with a reduction of serotonin (5- HT), norepinephrine (NE) and dopamine (DA) levels. DNA fragmentation by comet assay and histopathological analysis of cerebellum region of the brain were also evaluated. The results revealed that the selected drugs improved all parameters by variable degrees with the combination of coenzyme Q10 and niacin providing the most potent effect. In conclusion, coenzyme Q 10 and niacin are promising dietary supplements in the management of diabetic encephalopathy
