Abstract

This study was conducted to evaluate bacterial leakage of root canal obturation by System B and Endo Twinn and to evaluate adaptation of root canal filling at different canal levels by scanning electron microscope. Fifty extracted teeth with single root canal were and in this study. The teeth were prepared in crown down manner and irrigated by NaOcl 2.5%. The teeth were divided into three groups according to method of obturation group I(n=20), teeth obturated by using System B, group II(n=20), teeth obturated by using Endo Twinn, group III teeth obturated by lateral condensation. 7 days after complete setting of the sealer, groups were divided into sub groups according to aging time. Subgroup (A) was stored for 30 days, subgroup (B) was stored for 60 days. Each experimental group was inserted into tubes to be contaminated with bacteria, any test apparatus that showed sign of turbidity in the Brain Heart Infusion was discarded. Entercoccusfaecalis was cultured at 37oc for 24 hours in BHI then started to count bacteria by colony forming units (CFU)/ml. After bacterial leakage test each sample was sectioned longitudinal and prepared for drying and then sprayed with a layer of gold and then examined under Scanning Electron Microscope. In the present study it was found that after 30 days group I showed the highest significant mean of bacterial count, and no statistically significant difference between the control groups (with or without sealer). After 60 days group I showed the highest mean of bacterial count, and no significant difference between control group and group II, was found under Scanning Electron microscope the apical segment of group I showed a significantly higher mean of leakage than group II, while the middle and cervical segments showed no statistically significant difference
