Abstract
This study aimed to develop an intravesical delivery system of quercetin for superficial bladder cancer management in order to enhance drug efficacy, provide a controlled release profile and prolong the residence time inside the bladder. For this purpose, solid lipid nanoparticles (SLNs), either uncoated or chitosan coated loaded with quercetin were prepared and characterized in terms of colloidal, morphological and thermal properties. Drug encapsulation efficiency and its release behaviour were estimated. Furthermore, cytotoxicity of SLNs on T-24 cells was evaluated. Spherical SLNs (250 nm) ensured high entrapment efficiencies (EE > 97%) and controlled release behaviour over 142 h. In addition, cytotoxicity profile of SLNs revealed concentration-dependent toxicity recording an IC50 in the range of 1.6-8.9 μg/ qu r t . SLNs were further dispersed in an in-situ hydrogel comprising poloxamer 407 (20%) with either carbapol (1%) or chitosan (1%) as mucoadhesive polymers.
