Abstract

Background: Acute liver injury disorder contributes to liver fibrosis and cirrhosis which is very dangerous that may lead to hepatic cellular carcinoma (HCC) which is a common cancer worldwide. Stem cell-based therapy is considered a key area of research in end stage liver injury & diseases. Aim: This study aimed to elucidate the efficiency of different techniques to isolate non-parenchymal liver cells (NPCs) that contain stem cells from liver tissue through differential centrifugation and density gradient centrifugation, to characterize liver stem cells by flow cytometry using CD44& CD90, and, to evaluate the expression of Ý-catenin, Smad 2,3,7 genes in liver tissue using RT-PCR and to study the effect of curcumin and taurine on liver stem cells. Results: Cultured liver stem cells showed epithelial-like morphology at day 5-14. Liver stem cell markers significantly increased at day 12 and they were significantly higher in injured stem cells than non-injured-derived stem cells. Ý-catenin and Smad 2,3,7gene showed positive expression in injured liver tissues while no expression was detected in non-injured liver tissues. After curcumin and taurine treatments (5 , 400oM) for 24 h, a significant decrease in both CD90 and CD44 expression in injured-derived stem liver cells, whereas non-significant change was detected in the expression of both markers in non-injured-derived liver stem cells
