Abstract
Chapter 1: Formulation and Evaluation of Transdermal Elastic Nanovesicles Containing Rosuvastatin Calcium. Rosuvastatin calcium (RC) is mainly used for the treatment of hypercholesterolemia. However, its inhibitory action on HMG-CoA reductase also results in potential anticancer effect. The aim of this study was to develop and optimize novel elastic nanovesicular gel containing RC. This gel could act as drug carrier. Elastic nanovesicles (ENs) were prepared using thin film hydration technique using Tween® 80 (T80), cetyl alcohol (CA) and clove oil (Co). The OEND was converted into optimized EN gel (OENG) by using2% of Carboxy methyl cellulose (CMC). Results showed that the permeation extent from both optimized dispersion and gel through skin membrane was significantly higher than that from the drug suspension and the drug gel, respectively. Cell viability assay of the OENG on MCF-7 and Hela cell lines showed significant anti-proliferative and the potent cytotoxic effect when compared to the drug gel. Upon in vivo investigation in rabbits, the results have shown that OENG achieved increasing RC{u2019}s bioavailability in comparison with the drug gel. Finally, EN gel could be considered as promising nanocarrier for transdermal drug delivery of RC to increase its systemic bioavailability and potential anticancer therapeutic effect.
