Abstract

Agaricus bisporus mushroom is nowadays one of the most popular mushrooms around the world, not just because of its delicious taste, but also because of its bioactive compounds and their pharmaceutical values. The current work aims to induce the exopolysaccharides (EPS) production from Agaricus bisporus using different substrate additives, and study its effect as anticancer, antimicrobial and antioxidants substance. Different additives (tryptone, peptone, yeast extract, MgSO4, glucose, lactose, and sucrose) were tested individually using submerged culture for production of mycelia and exopolysaccharides. While tryptone produced the highest amount of mycelia biomass among others, sucrose produced the highest quantity of exopolysaccharides, which is then replaced with molasses by-product as a cheap available natural source of sucrose. Polysaccharide was then applied on MCF-7 cell line, showed no cytotoxic effect, while showed 30% time-independent antiproliferation activity. Also, q-PCR was used for evaluation of different apoptotic gene expression; TNFÜ, P53, Caspase9, BCL2, and Bax, in which TNFÜ showed the highest expression level (2.18 folds) after treatment. Assessment of apoptosis and cell cycle arresting using Flowcytometry, revealed that polysaccharides arrest G1/G2 phase, resulting in 5.5%apoptosis in treated cells
