Abstract

The objective of this thesis was to develop nano-systems as ocular delivery systems for Sertaconazole nitrate, that can overcome its poor solubility and would provide ease of delivery, acceptable ocular tolerance, stability, and enhanced transcorneal permeation. Hence, the work in this thesis was divided into the following chapters: Chapter I: Formulation and in-vitro evaluation of Sertaconazole nitrate loaded mixed micelles (STZ-MMs). Chapter II: Formulation and in-vitro evaluation of Sertaconazole nitrate loaded cubosomes (STZ-CUBs). Chapter III: Ex-vivo permeation study and in-vivo evaluation of the selected optimized Sertaconazole nitrate loaded formulations. from these chapters, it could be concluded that each of the optimized formulations, namely, Sertaconazole nitrate loaded mixed micelles formulations (MM-8 and MM-11) belonging to chapter I and Sertaconazole nitrate loaded cubosomal formulation (CUB-opt) belonging to chapter II, demonstrated promising mucoadhesive properties and good storage stability. The ex vivo corneal permeation study revealed that each of them significantly enhanced the transcorneal permeability of Sertaconazole nitrate. Moreover, each of them achieved good in vivo corneal tolerability and superior in vivo corneal uptake. Thus, they all can be considered as promising ocular delivery systems for treating fungal keratitis. Key words: sertaconazole nitrate, mixed micelles, cubosomes, confocal laser scanning
