Abstract

● Pasteurella multocida serotype 3 (P-1059) was cultured on brain heart infusion broth and then sub-cultured on brain heart infusion agar plates and, colonies appeared as circular (2–3 mm), smooth, convex, translucent, glistening, and butyrous, and show a tendency to coalesce.
● In Giemsa-stained films of Pasteurella multocida serotype 3 (P-1059), the organism appeared as rods occurring singly, in pairs, and occasionally as chains or filaments and bipolarity was demonstrated.
● Inoculation of Pasteurella multocida into embryonating chicken eggs via allantoic sac, resulted into embryonic mortalities 3-5 days post inoculation. Examination of embryos and embryonic fluids pointed out that dead embryos with hemorrhage and edema and clotted blood in embryonic fluids.
● Concentration of Pasteurella multocida serotype 3 (P-1059) in embryonic fluids were adjusted to be 107 CFU/Cm by colony forming unit technique.
● Inactivated chicken embryo derived bacterin was prepared by addition of formalin (0.3%) to allantoic fluid and then sterility was checked by culturing on brain heart infusion plate agar. Equal amount of adjuvant was added to inactivated allantoic fluid and checked for sterility.
● Results of our experimental infection revealed that clinical signs appeared 3 days post infection, sudden death occurred in non-vaccinated chickens of groups five and six. Other clinical sings include depression, decreased food and water intakes, whitish diarrhea and respiratory symptoms as cough, rales, nasal and ocular discharges, and facial edema.
● Postmortem examination of dead chickens showed that, some birds dead fleshy and in good condition, there was generalized congestion all-over the body. Internal examination reveals septicemic picture, liver and spleen were enlarged, congested and hemorrhagic, kidneys were swollen and congested, also there was congestion and exudates in trachea and bronchi, lungs were edematous and congested, intestine showing enteritis and pancreas was enlarged, congested and hemorrhagic.
● Mortalities, clinical signs, and postmortem lesions varied according to experimental infection and vaccination. Mortality was high in non-vaccinated chickens (groups five and six) and low in vaccinated birds (groups one to four). Clinical signs and postmortem lesions were severe in non-vaccinated birds and little to moderate in vaccinated birds. Chickens of control group showed no mortalities and no clinical signs.
● Results of SDS-PAGE revealed that Pasteurella multocida serotype 3 (P-1059) and Pasteurella multocida serotype 4 (P-1662) which were grown in chicken embryonating eggs via allantoic and yolk sacs inoculation, they showed specific bands 35Kd and 39Kd. While Pasteurella multocida serotype 3 (P-1059) and Pasteurella multocida serotype 4 (P-1662) which were grown on brain heart infusion broth and aga, they did not show specific bands.
