Abstract

Non-Steroidal Anti-Inflammatory Drugs (Nsaids)-Induced Gastric Ulcers Represent A Significant Clinical Concern And Adversely Affect Quality Of Life. Asymmetric Dimethylarginine/ Dimethylarginine Dimethylaminohydrolase-1 (ADMA/DDAH-1), Angiotensin Converting Enzyme-2/ Angiotensin 1-7 [ACE-2/ANG-(1-7)] And Inducible Nitric Oxide Synthase/Endothelial Nitric Oxide Synthase (Inos/Enos) Signaling Play A Crucial Role In Gastric Ulcer Pathogenesis.
The Present Study Was Planned To Evaluate The Role Of ADMA/DDAH-1, ACE-2/ANG-(1-7) And Inos/Enos Signaling In The Gastro-Protective Effect Of Perindopril And Tadalafil Against Indomethacin-Induced Gastric Ulcer In Rats By Comparing These Effects With The Standard Anti-Ulcer Drug, Pantoprazole.
Rats Were Randomly Divided Into 7 Groups Including; Normal Control, Perindopril (4 Mg/Kg/Day, P.O.), Tadalafil (10 Mg/Kg/Day, P.O), Indomethacin (60 Mg/Kg, Single Oral Dose), Indomethacin + Pantoprazole (40 Mg/Kg/Day, P.O), Indomethacin + Perindopril (4 Mg/Kg/Day, P.O), Indomethacin + Tadalafil (10 Mg/Kg/Day, P.O).
The Results Indicated That Pretreatment With Perindopril And Tadalafil Significantly Reduced Ulcer Index (UI) And Increased Preventive Index (PI) Together With Counteracting Indomethacin-Induced Histopathological Aberrations. Both Perindopril And Tadalafil Significantly Reduced Gastric Content Of Nitric Oxide (NO) While Significantly Increased Gastric Content Of Glutathione (GSH) And Superoxide Dismutase (SOD) Activity. In Addition, Perindopril And Tadalafil Significantly Reduced Gastric Tumor Necrosis Factor-Alpha (TNF-Α) And ADMA Expression While Significantly Increased Gastric Cyclooxygenase-2 (COX-2), Prostaglandin E 2 (PGE-2) And DDAH-1. Pretreatment With Perindopril Significantly Up-Regulated ANG-(1-7) Level And ACE-2 Protein Expression Measured By Western Blot Analysis. On The Other Side, Western Blot Analysis Revealed That Pretreatment With Tadalafil Significantly Reduced Inos Protein Expression While Enos Protein Expression Was Significantly Elevated.
It Could Be Concluded That Perindopril And Tadalafil Protect Gastric Mucosa from The Ulcerative Effects Of Indomethacin Through Exerting Potent Antioxidant And Anti-Inflammatory Effects Through Modulation Of ADMA/DDAH-1, ACE-2/ANG-(1-7) For Perindopril And Inos/Enos For Tadalafil.
