Abstract

This study had been carried out at the Department of Ornamental Horticulture, Faculty of Agriculture, Cairo University, Giza, Egypt, during the period from 2016 up to 2020, the experiments were done at tissue culture laboratory of Pico Modern Agriculture Company, Mansoriya, Giza, Egypt. The aim of this study was to establish a micropropagation protocol for the explants sprouted on the rhizomes of Calathea roseopicta ’Medallion’. Several sterilization techniques and culture media supplemented with different growth regulators were investigated on survival percentage, and the induction of shoots and roots of Calathea roseopicta ’Medallion’ plantlets. The obtained results showed that using [NaOCl (30% v/v) + ethanol (70% v/v) + HgCl2 (1 g/l w/v)] resulted in the most successful value (63.3%). Also, the maximum number of multiple shoots (21.2) lateral buds/shoot was obtained on MS medium supplemented with 1.5 (mg/L) 6-Benzyl adenine (BA) + 1.5 (mg/L) N6- furfuryl adenine (KIN) + 1 (mg/L) 1-Naphthalene acetic acid (NAA) + 30 (g/L) sucrose + 6 (g/L) agar. Test the effects of activated charcoal on the maximum roots induction (3.48 roots/shoot) was recorded on MS medium supplemented with 1 (mg/L) Naphthalene acetic acid (NAA) + 30 (g/L) sucrose + 6 (g/L) agar + 1 (g/L) activated charcoal, the plantlets were successfully acclimatized under greenhouse conditions
