Abstract

Monosodium glutamate (MSG) recorded one of the greatest food additives used all over the world. It improves the flavor of the food. A lot of studies founded that there are unbeneficial impacts with long time treated with MSG
Moreover, curcumin used as one of the strongest antioxidant compound and used nanoparticles of it improves the properties for curcumin.
The present study aims to examine the protective and therapeutic role of Nan-Cur versus monosodium glutamate toxicity in adult male albino rats.
A total number of 50 male rats weighing 120-180 g were used in the present study. The animals were divided into 5 groups (10 rats each).
• The first group served as control, received distilled water by gavage.
• The second group was treated with Nan-Cur (100mg/kg/ b.wt day).
• The third group was treated with MSG at dose of 10 mg/ kg b.wt/day.
• The fourth group was treated with Nan-Cur primarily for 4 weeks followed by Nan-Cur + (MSG) for another 4 weeks at the same doses.
• The fifth group was treated with MSG for 4 weeks followed by Nan-Cur for another four weeks at the same doses.
All control and treated groups were weighed and sacrificed at 4 and 8 weeks. The blood and tissue samples were collected for biochemical analysis.
The following parameters were evaluated
I -Lipid profile: through determination of total cholesterol (T-chol), triglycerides (TG), HDL, LDL.
II- Liver function testes: through determination of (ALT), (AST), total protein (TP) and albumin.
III- Oxidative stress: through determination of lipid peroxidation [(Malondialdehyde:(MDA)] and total antioxidant capacity (TAC).
IV-Heart enzymes and cardiac biochemical markers: through determination of serum lactate dehydrogenase activity (LDH), serum creatine kinase activity (CK), heart type fatty acid binding protein (H-FABP) and serum endothelial-1(Endo-1) level.
V- DNA fragmentation: through comet assay.
The results indicated that:
No significant difference was observed between control and Nan-Cur groups in most of the studied parameters.
• Lipid profile:
There was increase in the level of T-chol, TG, LDL while HDL level was decreased in MSG rats group. A marked decrease occurred in lipid profile levels in the protected rats group. A partial improvement in lipid profile levels in the therapeutic rats group. It is clear from the data recorded that the best improvement occurred in the protected rats group.
• Liver function testes:
There was an elevation was realized in ALT and AST activities in MSG treated group and improvement was observed in protected rats group.
A significant decrease in both total protein and albumin levels was reported in rats treated with MSG for 8 weeks. Furthermore, a slight decrease observed in serum total protein level in the protected rats group and an increase in serum albumin level at the last interval (8 weeks). Moreover, partial recovery occurred in the therapeutic rats group in total protein and albumin level respectively after 8 weeks of treatment.
• Oxidative stress parameters:
A significant elevation in MDA level was reported in rats treated with MSG for 8 weeks, a slight decrease took place in tissue MDA level in the therapeutic rats group and partial recovery occurred in the protected rats group.
A significant decrease in TAC level was reported in rats treated with MSG, slight decrease observed in therapeutic group and partial recovery occurred in the protected rats group.
• Heart enzymes and cardiac biochemical markers:
No remarkable changes in LDH activity were reported in protective rats group, in MSG rats group a significant increase in the activity of LDH was recorded and a significant decrease in LDH level was noticed in therapeutic group.
An increase was observed in CK activity in MSG rats group, partial recovery occurred in the protected rats group and slightly decrease in therapeutic group.
A significant altitude was noticed in H-FABP levels in MSG group as compared with control group and improvement was observed in protected rats group. There was a significant elevation was realized in Endo-1 level as compared with control group.
• Comet assay
As mentioned in the data of treated rats in protective group were nearly similar to that of the control group.
Conclusions:
In the current study we can included that Nan-Cur has the ability to improve the toxicity of MSG in liver and heart injury, we can take into consideration that Nano- particles(Nan-Cur) played a vital role as protective agent versus MSG toxicity dependent on time of treatment but its therapeutic potential was limited.
