Abstract

This dissertation consist of two parts: Part I In this part NiNPs/PEBT/GC modified electrode was fabricated by electropolymerization of eriochrome black T, immersion in 1 M NiSO4.6H2O solution and finally polarization for 35 cycle in 0.1 M NaOH. The study showed NiNPs/PEBT/GC modified electrode exhibited highly electrocatalytic activity towards the oxidation of glycerol (GLY) and stryptomycin (STRP). It can be used for detection of GLY and STRP and help us to study STRP-DNA interaction. Part II In this part we studied pentamidine (PNT) interaction with salmon sperm double stranded deoxyribonucleic acid (salmon sperm dsDNA) using a number of electrochemical approaches. The PNT-dsDNA interaction was investigated in solution and at a dsDNA biosensor surface. The peak currents of guanine and adenine were used as an evidence for the interaction of PNT drug with dsDNA. A simple electrografting process on GCE resulted in diazonized platform for covalent binding of DNA to the electrode surface. The interaction of PTN at the DNA modified surface was identified simply by CV measuring in CVI redox probe which binds preferentially to DNA. The interactions were further investigated by Faradaic EIS and CV in presence of ferri/ferrocyanide redox probe. The EIS proved to be an efective strategy for probing the bio-recognition events.
