Abstract

In the past two decades, dental materials have been widely improved
and along with the great development in tissue engineering and stem cell
biology led to open new era toward dental tissue regeneration and
differentiation.
The aim of our study was to evaluate and compare the effect of
mineral trioxide aggregate (MTA) and tri-calcium silicate cement
(Biodentine) on proliferation and differentiation of dental pulp stem cell
(In vitro study) and to evaluate and compare their effects on root
regeneration in immature necrotic permanent teeth. (In vivo study).
Pulp tissues of human vital impacted third molar teeth were removed
under sterile conditions and populations of dental pulp cells were isolated
using enzyme-digestion method, cultured and expanded in-vitro in culture
growth medium. The cultured dental pulp stem cells were characterized
by monitoring their growth, self-renewal capability, Multidifferentiation
lineages and colony formation during the culture period. Also, the relative
expression of stem cell genes (Bmi1 & Stat3) in the cultured cells was
evaluated using quantitative Reverse Transcription Polymerase Chain
Reaction (RT-PCR).
