Abstract

Cancer is considered one of the most common causes of morbidity and mortality worldwide. Many research focused on discovery of natural compounds that can be used in the prevention and/or treatment of cancer as many chemotherapeutic agents have been associated with many side effects. Ehrlich ascites carcinoma (EAC) model was used in several studies that indicate the effectiveness of some chemotherapy and plant sources against cancer, this is because of the similarity with human tumors in the fast of growth and undifferentiated and their sensitivity to chemotherapy. The present study was undertaken to examine the therapeutic and antineoplastic effects of Cisplatin, Oxaliplatin and vitamin B17 against Ehrlich ascites carcinoma–induced in female albino rats. The animals were randomly distributed into seven groups, each group contain seven female rats as follows: Normal control rats (group A). Positive control (group B) rats that injected by EAC as a cancer model. group C, Rats that injected by EAC and treated by Cisplatin. group D, Rats that injected by EAC and treated by single dose of Oxaliplatin. group E Rats that injected by EAC and treated by Vitamin B 17. group F, Rats that injected by EAC and treated by Cisplatin plus Vitamin B 17. group G, Rats that injected by EAC and treated by single dose Oxaliplatin plus Vitamin B 17. After one week from beginning of drugs treatment, blood samples are collected and Tumor markers AFP,CEA ,CA 19-9, TPA and LDH as well as Antioxidants bio enzymes SOD,CAT,GSH and MDA were measured .Complete Blood Count, Blood Glucose, Serum Amylase, ALT,AST,ALP, Serum Albumin, Total Bilirubin, Serum Creatinine, BUN levels were estimated, histopathological examination of liver, kidney and spleen with Immunohistochemistry (P53, Bcl-2 and PCNA) were assessed for study the antitumor activity and side effects of drugs used on hepatic and renal tissue. Results showed that the treatment of VB17 alone or in a combination with Cisplatin and Oxaliplatin lead to decrease AFP, CEA, CA 19-9, TPA and LDH when compared with (EAC) - bearing rats. Reduced levels of MDA with enhanced levels of antioxidant indicators were observed in the Vitamin B17 group alone or in a combination with Cisplatin and Oxaliplatin. Statistical analysis showed significant increase in the activity of enzymes (ALT, AST, ALP) accompanied by an increase in the levels of Total Bilirubin, Serum Creatinine and BUN in the treated groups by Cisplatin and Oxaliplatin which approve their toxic effects on Hepatocytes and renal cells which confirmed with histopathological examination and Immunohistochemistry
while VB17 groups show improvement in previous parameters. It could be concluded that Vitamin B17 has possess anticancer and antioxidant activities that justify its traditional use, and its potential hepatoprotective effect and kidney ameliorative role.
