Abstract Although the enormous previous research discussed the variable pharmacological activities of dihydropyridines (DHPs), none has designated the full detailed mechanism of action at the molecular level of this nucleus Hence our research focused on three major biological activities anticonvulsant, antidiabetic and anticancer effects to dig deeply at molecular levels how DHPs exert their clouts and reveal answers around their mechanism Numerous questions have been needed to be addressed, also the role of calcium channel blocking in the diverse biological effects of DHPs. To answer all these questions, we synthesized 21 novel DHPs with different side chains and aldehydic-based cores to evaluate their biological actions extensively via in vivo, in vitro, cell lines and enzymatic assays. Moreover, biochemical parameters and histopathological examination were used. Furthermore limited structure activity relationship depending on structural variations was conducted to develop a paradigm for future drug discovery based on our achievement and open the gates for repositioning DHP nucleus in new druggable areas. |
