Abstract Nitrosodiethylamine (NDEA) induces hepatocellular carcinoma in rats. This study aimed to estimate the possible therapeutic role of metformin in N-nitrosodiethylamine induced hepatocellular carcinoma. Fifty male albino rats were used in this study, divided into five groups includes (control, metformin, NDEA, protected and treated groups). Serum liver function was estimated. Markers of oxidant stress and antioxidant markers as glutathione peroxidase, superoxide dismutase, reduced glutathione, vitamin A and vitamin E were estimated. In addition, Bcl-2 and caspase-3 were estimated in addition to histopathological examination. Liver Fe, Cu, Zn were also estimated. The data showed that NDEA increase liver enzymes, oxidant parameters, Bcl-2 expression and decrease liver protein, albumin, antioxidant parameters and caspase3. Contrarily metformin improved these values in protected and treated groups. In conclusion our results revealed that; the protective effect of metformin on hepatocellular carcinoma is better than its treated effect, so metformin might be used as protective agent in HCC. This will open new perspectives that Metformin is a protective compound to prevent, slow or treat the occurrence of liver cancer.
