Abstract Model organism C.elegans has seven homologs of cytochrome b561. Among them, Cecytb-2 was confirmed to be expressed in digestive organs.Cecytb-2 was expressed in Pichia pastoris cells, purified and then assembled into phospholipid bilayer nanodiscs. The reconstituted Cecytb-2 was more stable and more reducible by ascorbate than detergent-solubilized ones. We measured the oxidation of reduced heme of Cecytb-2 upon addition of ferric substrates under anaerobic condition in both detergent micelle and nanodisc states. The putative ferric reductase activities in both states showed clear dependencies on the substrate concentrations.Further, we confirmed that the ferric substrates were converted to ferrous state by nitroso-PSAP assay. Importantly, we observed that the ferric reductase activity of Cecytb- became enhanced in the phospholipid bilayer nanodisc |
