Abstract This thesis is devoted to the development of new analytical methods for the estimation of some CNS acting drugs ; namely clozapine (CLZ), caffeine (CAF), ergotamine tartrate (EGT) and phenytoin sodium (PHY). This thesis consists of four main parts: Part I : General introduction about the studied drugs including : pharmacological action, chemical structure, physical and chemical properties and literature review.Part II : this part is divided into two subdivision Part II A : Represent a simple and validated spectrophotometric method have been for the measurement of clozapine (CLZ) depending on its interaction with N-bromosuccinimde (NBS) resulting in formation of a yellowish orange colored product, measured at 320 nm. Part II B : Introduce a colorimetric method which depends on the condensation of the CLZ with acetic acid mixed anhydride reagent producing a purple colored product measured at 319 nm. The linearity range was 8.0-24.0 µg/mL. Part III : In this part, a new, sensitive and reproducible hybrid micellar liquid chromatographic method for the quantification and separation of CLZ and PHY in pure, dosage form and in biological fluids was presented. The suggested method was subjected to detailed validation parameters according to the ICH guidelines.Part IV : This part aims to develop two simple, sensitive and validated methods for estimation of ternary mixture of ergotamine, caffeine and dipyrone, namely ; double divisor ratio spectra as a method A and ratio spectra first derivative spectrophotometric method as method B. Part V : This part presents the construction of a calixaren-8 modified ion selective membrane electrode for rapid determination of clozapine (CLZ) intoxication over a glassy carbon electrode using phosphotungestic acid as a cationic exchanger and nitro phenyl propyl ether as a plasticizer in a PVC based matrix. |
