Abstract

To investigate the effect of different antioxidants (cysteine and L-carnitine) and cryoprotectants (raffinose and trehalose) supplementation in semen extender on post-thaw buck semen parameters. Semen was collected from 5 bucks once weekly for 10 successive weeks during the breeding season. Semen was divided into 13 aliquots (control - 2.5, 5, 10 mM cysteine – 2.5, 5, 7.5 mM L-carnitine – 5, 10, 15 mM raffinose and 50, 75, 100 mM trehalose). Semen was cryopreserved and stored in liquid nitrogen tank. After thawing, semen was evaluated. The results showed that cysteine and Lcarnitine significantly (P˂0.001) improved the post-thaw sperm criteria, DNA integrity, diminishing the leakage of sperm enzymes with increase in levels of TAC and decreased lipoperoxidation production. The best results were obtained by concentrations of 10 mM cysteine and 7.5 mM L-carnitine. Also, raffinose and trehalose significantly (P˂0.001) improved sperm motility, viability, membrane and acrosome integrities. The best results were recorded at 5 and 15 mM raffinose and 50 mM trehalose. In conclusion addition of 10 mM cysteine and 7.5 mM L-carnitine and 5 and 15 mM raffinose and 50 mM trehalose to semen extender improved semen quality post-thawing.
