Abstract

The present study dealt with investigates of biochemical properties as follows The highest total polyphenol content was determined in marigold flowers EtOAc fraction followed by its methanolic extract (125,18 and 57,52mgGAE/g D.W, respectively. The highest content of total flavonoid was also measured in marigold flowers EtOAc fraction followed by its methanolic extract (112,81 and 67,88mgRE/g D.W), respectively. Contrary, mums flowers methanolic extract contained higher total flavonoid content than its EtOAc fraction (37,75and 34,82mgRE/g D.W), respectively Twenty nine bioactive compounds were identified using HPLC technique .The analysis cleared that acacetin neo-rutinoside and naringin were absence in mums green aerial parts methanolic extract. Beside this, coumarin wasn’t found in EtOAc fraction of mums flowers and green aerial parts. Marigold flowers methanolic extract contained the highest amount of identified flavonoid compound apig-6rhamnose-8-glucose (18,34mg/g extract).Additionally, salicylic acid and ellagic acid weren’t detected in the same extract. In vitro antioxidant activity evaluation proved the superiority of marigold flowers EtOAc compared with standard references with the lowest detected IC50value in DPPH assay 32,5µg/ml. The highest reducing power ability was found in mums flowers EtOAc fraction with ED50 value 25,19µg/ml. Correlation analysis between phytochemical compounds and antioxidant assays in mums flowers and green aerial parts proved that phenolic compounds were vital in DPPH assay and total antioxidant capacity. Correlation analysis between phytochemical compounds and antioxidant assays in marigold flowers and green aerial parts showed that phenolic and flavonoid compounds were dynamic in all applied antioxidant methods. In vitro antibacterial activity evaluation using three Gram-negative plant pathogenic bacteria showed that methanolic extracts were more active compared with their EtOAc fractions. The best antibacterial action was recorded against Bulkholderia cepacia followed by Pectobacterium carotovora using marigold flowers methanolic extract at a concentration of 1000mg/ml. The lowest detected antibacterial activity was noticed against Bulkholderia cepacia using mums green aerial parts EtOAc fraction. In vitro nematicidal activity against Meloidogyne incognita egg hatching was evaluated using three different concentrations. The best examined sample in ihibition Meloidogyne incognita egg hatching was marigold green aerial parts EtOAc fraction followed by its methanolic extract at a concentration of 1000µg/ml. Pot experiment was performed to evaluate the potentiality of marigold green aerial parts methanolic extract and its derived EtOAc fraction towards Meloidogyne incognita under in vivo conditions using green sweet pepper. The highest enzyme activities for peroxidase and polyphenol oxidase was noticed in the 6th day after inoculation with Meloidogyne incognita eggs + second stage juvenils. Their values were 166,19 and 85,87 U/g F.W/min, respectively. Nine weeks after cultivation some plant parameters were determined and all of them affirmed the excellence of marigold green aerial parts EtOAc fraction compared with its methanolic extract and control seedlings.
