Abstract

Peripheral neuropathy is one of the annoying side effects of cancer chemotherapy that obligate the patient to stop the drug or decrease its dose and this can deteriorate the case. It can occur with many drugs but vincristine which is one of vinca alkaloids is commonly used as a combination regimen for treatment of vast number of tumors and in pediatric tumors. It binds for B- tubulin which is important as a cytoskeletone for neuronal axon and mitosis but also it has other mechanisms of action.Pregablin is widely used as anticonvulsant drug and in treatment of neuropathic pain. It binds to α2Ϭ subunit and inhibits Ca influx so, the release of many chemical transmitters as (Noradrenaline, glutamate and substance B) leading to inhibition of pain. 40-60% of patients of neuropathy respond to treatment by pregablin so, other drugs should be added to improve the results but this increase side effects so, investigations should be done to discover neuroprotective drugs with new mechanisms and few side effects.Metformin is a classic medication for treatment of type 2 diabetes and has a protective role in diabetic neuropathy and this calls researches to study its role in chemotherapy induced prephiral neuropathy. It has antioxidant,anti-inflammatory, anti-apoptotic effects and helps development of nerves by production of NGF and has few side effects so, it is an ideal treatment for combination with pregablin
