Abstract

The present work was carried out on 40 male albino rats with body weight (200-250 gm). All animals were housed in isolated animal cages, under controlled environmental conditions, 12/12 h light/dark cycle, at room temperature (23 ± 2°C), with free access to standard rat chow and drinking water. All procedures were done according to the ethical committee of Faculty of Medicine, Tanta University. The animals were acclimatized for two weeks, then randomly divided into 4 groups; 10 rats per each: • group I (Normal control group):- The animals of this group received 0.5 ml physiological saline as a single dose intraperitoneally (i.p) for 9 days. • group II (Quercetin group):- The animals of this group received a daily dose of quercetin (50 mg/kg) orally for 9 days. • group III (Mtx treated group):- The animals of this group were treated with Mtx at a single i.p. dose of 20 mg/kg dissolved in 0.5 ml physiological saline at the 1st day. • group IV (Mtx + Quercetin treated group):- The animals of this group received a daily dose of quercetin (50 mg/kg orally) for 9 days, in addition to a single i.p. dose of Mtx (20 mg/kg dissolved in 0.5 ml physiological saline) administered 30 min. after the 3rd dose of quercetin. At the end of experiment, all animals will be scarified by cervical decapitation under general anesthesia to collect blood samples and take sample of renal tissue. For measuring the following parameters: -  Serum Urea.  Serum Creatinine.  Serum TNF alpha.  Renal Reduced Glutathione (GSH).  Renal Malondialdehyde (MDA). The achievement gained in this study could be summarized as follows: Quercetin group (II) showed insignificant changes in all studied parameters as compared to the Control group (I). While, the group treated with Mtx (III) showed significant increase in the serum urea level, serum creatinine level, serum TNF-α level and the level of renal MDA with significant decrease in the renal GSH level as compared to the Control group (I) and Quercetin group (II). However, the group recieved Mtx 20 mg/kg then treated with quercetin 50 mg/kg (IV) showed significant decrease in serum urea level, serum creatinine level, serum TNF-α level and the level of renal MDA with significant increase in the renal GSH level as compared to the group received Mtx alone (III).
