Abstract

Chronic B cell lymphocytic leukemia (CLL), arises from the bone marrow and is affecting the majority of body tissue. Here in this study, authors aimed to evaluate the level of interleukin (IL)-9 gene expression by quantitative reverse-transcriptase polymerase chain reaction (qRT-PCR) and its protein level by enzyme-linked immunosorbent assay (ELISA) from blood samples, the level of T helper (Th)9 cells by flow cytometry, and determination of the level of oxidative stress in CLL’s patients using spectrophotometric and native gel electrophoresis methods followed by assessment of the relation between the aforementioned parameters with the prognosis of patients with CLL. Current study involved 70 Egyptian patients with CLL who were admitted to the Oncology Center of Mansoura University, Mansoura, Egypt, between December, 2016 and July, 2017 in addition to 15 apparently healthy individuals were selected to act as a control reference group. The current study indicated that Th9 cells and IL-9 contribute to the pathogenesis of CLL and are correlated with TP53 Del (17p), BCL2 expression and the more complex form of disease and worse prognosis. Enhanced levels of oxidative stress (MDA and PC) and decreased levels of enzymatic anti-oxidants (SOD and catalase) in CLL patients reflect the pathological state and impaired disease control in CLL patients. Levels of Th9 cells and IL-9 were strongly correlated with oxidative stress and bad prognosis in patients, indicating that these cells may contribute to CLL by novel mechanisms that could include oxidant injury. These results may allow one to speculate a potential therapeutic target of Th9 cells and IL-9 in CLL.
