Abstract This study was carried out to detect the polymorphisms in GH gene in two duck breeds Muscovy and Pekin ducks by direct sequencing and single strand conformation polymorphism (SSCP). To achieve these objectives, the following methodology was used This work was conducted at the research unit of molecular biology and biotechnology, Animal Wealth Development Department, Faculty of Veterinary Medicine, Zagazig University. 1. Blood samples were collected from a total number of 112 mature healthy ducks (56 of them were Muscovy and the other 56 were Pekin ducks) and then DNA was extracted. 2. Three amplicons of growth hormone gene were amplified including: a. Amplicon I (part of 5` UTR, exon I and its partial flanking intron) of GH gene in Muscovy and Pekin duck breeds. b. Amplicon II (exon II and its partial flanking introns) of GH gene in Muscovy and Pekin duck breeds. c. Amplicon III (exon IV and its partial flanking intron) of GH gene in Muscovy and Pekin duck breeds. 3. SNPs were then detected using: single strand confirmation polymorphism (SSCP) followed by direct sequencing. Results obtained could be summarized in the following points I- Amplicon I (part of 5` UTR, exon I and its partial flanking intron) of GH gene in Muscovy and Pekin duck breeds: A product of 415 bp in size was extracted in both Muscovy and Pekin ducks under studying. |
