Abstract Amygdalin is considered one of the nitrilosides, natural cyanide containing substances abundant in the plants seeds of the prunasin family that have been used to treat cancers and also as pain reliever. Particularly, D-amygdalin (D-mandelonitrile-b-D-gentiobioside) known to exhibit apoptotic effect on cancer cells. It’s used as antitumor drug. If it combines with conditional chemotherapy drugs it exhibit synergistic effect. Amygdalin also has role in curing other diseases such as controlling asthma, improving immune system, inducing apoptosis of human renal fibroblast, inhibit hyperglycemia. The aim of the current study is to examine the anti-tumor effect of extracted amygdalin and pure amygdalin on the MCF-7 breast cancer cells, so we applied ethanolic extraction of amygdalin from bitter almond, sweet apricot and cashew seeds and application a high performance liquid chromatographic procedure for amygdalin quantification to evaluate extraction efficiency and evaluate if amygdalin induces cell death on human breast cancer cells MCF-7 by Bax, caspase-3 activation and decrease of Bcl-2 expression. After statistical analysis our results referred to the following: Amygdalin analysis by HPLC It was necessary to establish a suitable HPLC method to determine amygdalin in the seeds. Agilent C18 column and methanol–water as mobile phase were used. It was found that when the ratio of methanol and water was 15:85, amygdalin could be completely separated with the other ingredients. |
