Abstract

The present study investigates some pharmacodynamics effects of fenbendazole and rafoxanide and their concomitant administration in mature male rats.
Experiment on mature rats
Mature mole rats were divided into 4 groups each group of 15 rats.
Group1: Rats were treated with fenbendazol (37.5 mg/kg. b.wt. orally).
Group2:Rats were treated with rafoxanide (7.5mg/kg .b. wt. orally).
Group3: Rats were treated with fenbendaxole concomitantly with rafoxanide.
Group4: Rats not treated by any drugs and kept as control group.
Blood samples were collected from treated and control rats at the end of 2nd, 4th, 8th week from drug administration to examine the effect of these drugs on blood picture, hepatic and renal functions, semencharacter, Index weight of some body organs and the histological structure of reproductive organs, liver and kidney.
The obtained results are as follows:
1- Finding on mature male rats:
The effect of fenbendazole (37.5mg/kg. b. wt. orally) or rafoxanide (7.5mg/kg. b. wt. orally) each alone and their concomitant administration on:
A- Hematological pictures:
Hb concentration showed a significant decrease at all periods of treatment with fenbendazole, also showing significant decrease at 4th and 8th after administ ofrafoxanide a lone and fenbedazole with raf (Concomitantly) respectively.
PCVShowed significant decrease ingroup treated with fenbendazole allover periods of experiment week of rafoxanide and 8th week in group treated withconcomitant drugs.
Male rat treated with rafoxanidesshowed increase in PCV after 2nd week, also there was significant at 2nd and 4th weak.
RBCsCount showed signficant. Decrease at 2nd, 4th weak after administration of fenbendazole and showed song increase in 8th week.
RBCsIn rafoxanide treated group decrease 4th and increase 8 W.
RBCs In concomitant treated group was found increase at 4th weak.
WBCs significant increase in all treated group all over the period of the experiment.
B-Biochemical finding:
1- Liver function tests:
ALT:Showed a signification increase from treatment with rafoxanide.
AST:Showed asignificant increase in all treated group all over the periods of the experiment.
ATP: Showed a significant increase in all treated group all over the periods of the experiment.
Total protein: Showed a significant decrease at 4th week after administration of rafoxanide and also showed a significant decrease in albumen level at 4th weak from treatment.
Globulin level showed significant increase at 4th week after treatment with fenbendazile alone and concomitant drug, there was also significant decrease after 2nd week of administration of concomitant drug.
Total cholesterol level showed significant increase in all treated groups all over the period of experiment.
Triglyceride level showed significant at 2nd and 8th weak after administration fenbendazole and rafoxanide each alone.
Concomitant group, showed significant increase after 4th and 8th week.
2- Kidney function tests:
Urea levels were increase significantly in all treated groups all over the periods of experiment.
C- Fertility finding
Sperm count showed a significant decrease at 2nd, 4th and 8th weak from treatment in all treated groups.
Sperm motility showed a significant increase in all treated groups all over the periods of the experiment.
D-Index weight of some internal organs:
Index weight of liver and kidney showed non-significant change, showed significant increase in Index weight of testes in all treated group.
E-Histopathological finding:
Male rats treated by alone and their concomitant administration showed many histological alterationsuch as liver showed congestion of hepatic blood vessel, degenerative glomeruli in kidney, testes showed marked degeneration of germinal epithelium with sloughing of testicular cells in the lumen, also epididymis revealed absence of spermatozoa.
