Abstract

L.arginine as substrate for NOS, usually not significantly improve the changes in most tested parameter when compared with Aβ and AlCl3-treated rats. That’s effects may be due to overproduction of NO.
2- L. NAME and 7-NI by inhibition of NOS, improved the disturbance occurring by Aβ and AlCl3.
3- Regulation of NO level is the key for stability and prevention of neurodegenerative effects. Over production of NO induce neurotoxicity related to AD.
4- Combination of Donepezil with L. NAME and 7-NI highly significant protect against Aβ and AlCl3 toxicity.
5- Aluminium chloride increase level NO, AChE, MDA and decrease level SOD, CAT, GSH and ALA more than Aβ. While, amyloid beta increase latency time in MWT, AA, LA, IL-β and TNF-α more than AlCl3 but bot induced neurotoxicity.
8- Recommendations
1- Patients with AD may not identify toxic substances, so caregivers should ensure that remove all harmful objects.
2- Taking Mediterranean diet, that associated with lower AD risk and lower mortality in AD. This diet is rich by anti-oxidants. And its including fish, vegetables, fruit, cereals, unsaturated fatty acids (e.g., olive oil)
3- Taking supplements containing antioxidant such as omega-3 and vitamin E.

4- Combining antioxidant and anti-inflammatory agents to the AD drugs is necessary.
5- Further investigations are necessary to explain the role of NO in AD. And possibility of addition of NOS inhibitors to AD treatment.
