Abstract Myocardial infarction (MI) represents a major health problem worldwide. Current study aimed to investigate the therapeutic angiogenic effect of α7-nAch receptor activation non-selectively by nicotine or selectively by PHA in MImodel in rats. MI was induced by isoprenaline. MI rats were treated with nicotine or PHAor MLA,followed by injection of nicotine one hour later, all treatments were given for 16 days. Histopathological examination of left ventricles was carried out. The plasma level of NO and VEGF was determined. α7-nAch receptor, iNOS, HO-1 and CD34 expression was elucidated by immune staining. Isoprenaline-insulted rats showed a significant ST segment elevation and prolonged QT interval. VEGF and NO plasma levels were elevated. Ischemia reduced the capillaries density that accompanied with diminution in α7-nAch receptor and INOS expression. The expression of HO-1 was enhanced. Activation of α7-nAch receptor increased the capillaries density and reversed the histopathological and ECG anomalies of MI. conversely, the expression of α7-nAch receptor was decreased leading to consequent reduction in VEGF and NO plasma level and iNOS expression. Furthermore, the recovery of cardiac tissue reduced HO-1 expression. |
