Abstract

This study aimed to evaluate IgG-binding to megakaryocytes in bone marrow specimens of ITP and MDS patients as a step towards determining megakaryocytes targeting by autoantibodies in vivo.
Subjects were classified into two groups:
group I: (ITP patients):
Included 20 patients diagnosed as primary ITP based on established clinical criteria, platelet count levels below 100 x109/L at the time of bone marrow sampling and no other pathology identified on bone marrow examination. This group included 15 females (75%) and 5 males (25%).Their ages ranged from 25 to 68 years with mean ±SD (36.9 ± 11.4).
group II: (MDS patients)
Included 20 thrombocytopenic patients whose clinical presentation and bone marrow features were consistent with myelodysplastic syndrome (MDS). They had platelet counts below 100 x109/L at the time of bone marrow sampling. This group included 11 females (55%) and 9 (45%) males. Their ages ranged from 38 to 71 years with mean ±SD (52.9 ± 8.7).
Both groups were subjected to the following:
1- Complete history taking:
Considering age, sex, fever, bleeding tendency, easy fatigability, liver disease and drug history
2- Clinical examination:
Including: pallor, purpura, liver, spleen and lymph node enlargements.
3- Laboratory investigations:
A) Routine investigations: for group I
1-Complete blood count (CBC)
2- Erythrocyte sedimentation rate (ESR)
3- Liver function and renal function tests
4-Viral markers (HBsAg,HCVAb,HIVAb)
5-Prothrombin concentration and INR
6. Autoimmune profile (ANA,RF,anti dsDNA).
