Abstract

The present study was conducted to record the possible adverse effects of Dexamethasone and the role of Silymarin in restoring or minimizing these effects in male albino rats. For these experiments 65 male rats were randomly divided into 4 groups, each of 15 rats except the 4th group which contain 20 rats as follows: group 1:(control group): Rats were daily given distilled water (2ml/kg. b.wt) orally by stomach tube for one month and saline (2ml/kg. b.wt.) intramuscularly twice weekly with interval of 3 days for one month. group 2: (Dexamethasone treated group): Rats were treated with Dexamethasone at a dose level of 0.25 mg/kg. b.wt i.m. twice weekly with interval of 3 days for one month. group 3:(Silymarin treated group): Rats were daily treated with a single dose of Silymarin at a dose level of 50 mg/kg b.wt. given orally by stomach tube for one month. group 4 :( Dexamethasone +Silymarin treated group): Twenty rats were treated with Dexamethasone for one month concomitantly with daily treatment with Silymarin for one month . Five rats from this group were treated with Silymarin alone for another one month. The duration of experiment extended to 2 months. Five rats from each treated and control groups were sacrificed after 2nd week, 1 month and 2 months from the beginning of drug administration . Blood, body organs and epididymal contents were obtained from all groups .All experimental rats were weighted before sacrificed . obtained results are summarized as follows: 1-Hematological Findings: Administration of Dexamethasone induced a significant reduction in R.B.Cs count,W.B.Cs count,PCV% ,eosinophil’s and lymphocytes count in male albino rats. Administration of Silymarin concomitantly with dexamethasone induced a significant increases in R.B.Cs count ,W.B.Cs count ,PCV% ,eosinophil’s and lymphocytes count in male albino rats. 2-Biochemical Findings : Dexamethasone induced a significant increase in serum levels of ALT, ALP , triglycerides ,urea and creatinine as well as it induced a significant decrease in serum total protein and globulin values in male albino rats. The drug induced a significant increase in MDA content and a significant decrease in total antioxidant capacity levels in serum. Administration of Silymarin concomitantly with Dexamethasone induced a marked improvement in all previous biochemical parameters induced by Dexamethasone in male albino rats. 3-Effects on index weight of some body organs : Dexamethasone induced a significant a increase in index weight of the liver and kidney at 1 month of experiment . Also, the drug induced a significant decrease in index weight of spleen at different periods in male albino rats. Administration of Silymarin concomitantly with Dexamethasone induced a marked improvement in index weights of these organs as compared with dexamethasone treated groups . 4-Effects on semen characters : Dexamethasone induced a significant increase in index weight of epididymis at 1 month and accessory sex glands at 1 and 2 months of the experiment. The drug induced a significant decrease in total sperm count and motility % as well as it induced a significant increase in total sperm abnormalities % in male albino rats. Administration of Silymarin concomitantly with Dexamethasone induced a marked improvement in all previous changes in semen characters induced by dexamethasone 5-Histo-pathological Findings: Repeated administration of Dexamethasone induced deleterious effects on some body organs as liver , kidney ,spleen ,lymph node, tests, epididymis ,accessory sex glands . Administration of Silymarin concomitantly with Dexamethasone succeeded to ameliorate these lesions in the treated male rats.
