Abstract

The present work aimed to the production of silver nanoparticles by the bio-reduction potential of phycocyanin extracted from the cyanobacterium Nostoc linckia as well as phycoerythrin extracted from the cyanobacterium Nostoc carneum as non-toxic eco-friendly biological method. The production of silver nanoparticles (AgNPs) were statistically optimized by detection of the most suitable chemical and physical variables to obtain the maximum production of AgNPs, using different bio-reduction parameters as well as the properties of AgNPs were also investigated. AgNPs showed antimicrobial activity against S. aureus, E.coli, Pseudomonas aeruginosa, Klebsiella pneumonia, Streptococcus sp. and Enterobacter aerogenes. The biosynthesized AgNPs are found to be effective in resisting the action of the complete hemolysis caused by AAPH free radicals. The anti-hemolytic percentage of AgNPs of phycocyanin was 92.2 % and 96.2 % in case of AgNPs of phycoerythrin. Concerning anticancer activity, AgNPs exhibited higher cytotoxic effect against mammary gland breast cancer (MCF-7) cell lines than the normal cells, human lung fibroblast (WI38) and human amnion (WISH) cell lines (in vitro). AgNPs have induced growth inhibition of Ehrlich ascites carcinoma (EAC) cell lines in the experimental animals (in vivo). Treatment with AgNPs at the dose of 5 mg/kg body weight increased the hemoglobin content and RBC count which is higher than treatment with 5-FU (antibiotic). Also the total WBC counts, tumor cell count and tumor volume were found to be decreased after treatment with AgNPs which is lower than treatment with 5-FU.
