Abstract Three experiments were carried out to investigate the effect of Three types of continuous wave (CW) laser irradiations,vis.diode, 650 nm (50mw), 2nd harmonic of Nd: YAG, 532 nm (100mw) and NIR diode,808 nm (2 W)on the motility, viability, acrosome reaction, membrane integrity(AST, ALT and ALP), as well as DNA integrity of human spermatozoa. Semen samples were allowed to liquefy for 30 min at 37°C to evaluate the seminal parameters before and after laser exposure. The experiment was repeated three times for each treatment (treated sperm population). Semen samples were conventional analyzed just after liquefaction, then diluted 1:5 in S.TALP buffer. Diluted samples were split into eleven portions of 0.5 ml each. Ten portions were irradiated by three different lasers for different exposure times that vary from 1 to 10 min., while the eleven one was control sample (initial sperm population). Sperm motility, Sperm abnormalities, percentages of live spermatozoa and acrosome reaction were assessed. Determination of AST, ALT and ALP activities were performed by colorimetric method. Assessment of sperm DNA integrity, tail length and tail moment were carried out using single cell gel electrophoresis (SCGE), comet assay. |
