Abstract
The present study was carried out to evaluate some pharmacodynamic properties of Enrofloxacine and ampicillin each alone and their concomitant administration to male rats. The possible adverse effects of these drugs were studied. The experiments were carried out on 60 mature male albino rats of 150-180 gm. body weight each and about 140 – 160 days old . For this experiment mature male rats were divided equally into 4 groups each of 15 rats as follows:
group I: Rats were injected with distilled water at a dose of 2ml/kg b.w. s/c for 5 successive days and kept as a control group.
group II: Rats were injected with ampicillin ( 40 mg/ kg b.w. s/c twice daily for 5 successive days ).
group III: Rats were injected with enrofloxacine( 18 mg / kg b.w. s/c once daily for 5 successive days ).
group IV: Rats were injected with both enrofloxacine ( 13.5 mg/kg b.w . s/c) once daily + ampicillin(30 mg/kg b.w. s/c ) twice daily for 5 successive days .
Five rats from each treated and control groups were killed after 2nd,4th and 8th week from the beginning of drug administration . Blood ,body organs and epididymal contents were obtained from treated and control rats .
The obtained results are summarized as follows :-
1- Effects on blood picture :
There was a significant decrease of W.B.Cs. count in rats treated with Ampicillin after 2 and 4 weeks from drug administration and a significant reduction of R.B.Cs. in rats treated with Enrofloxacine after 4 and 8 weeks. Also, there was a decrease in Hb value rats treated with Ampicillin and Enrofloxacine at 4th week . Moreover there was a reduction in P.C.V. value in all treated rats at allover the experimental periods .
2- Effects on some biochemical parameters :
The obtained data showed that there was a significant increase in serum ALT & AST values in rats treated with Enrofloxacine at 4th weeks of the experiment. There was an increase of ALP level in all treated rats at 4th and 8th week .
There were insignificant changes in level of serum protein level and serum albumin level of all treated rats allover periods of experiment .
Also, there was a significant increase serum urea in all treated groups at the 8thweek of the experiment and there was elevation in creatinine level in all treated rats at allover experimental periods .
3-Effects on some organs weight :
There were significant changes in some internal organs weight ( liver ,kidney ,testes, epididymis and accessory sex glands ) in treated rats .
4- Effects on semen characters :
The obtained results showed that administration of Ampicillin induced a reduction in the progressive sperm motility and sperm count values at 2nd week of the experiment. Also , there were non significant changes in sperm motility of mature rats in other groups after 2, 4 and 8 weeks from drug administration . However, there was an increase in sperm abnormalities in all treated groups at the 2nd week of the experiment .
5-Histopathological findings :
Histo- pathological examination of the liver , kidney and the reproductive organs isolated from male rats revealed some histological alterations at different periods of the experiment. .
