Abstract

The current study was delineated to evaluate the possible hematoprotective and immunomodulatory effect of vitamin C and E. Purpurea in cyclophosphamide treated mice. For this purpose, sixty four adult male albino mice were randomly allocated into eight equal groups.
The results revealed that:
I- Effect of Vit C and/or Echinacea Purpurea on hematological parameters of cyclophosphamide treated mice.
A) Effect on Erythrogram: Treatment with CYC alone once daily at a dose of (25 mg/kg b.wt.) for ten successive days provoked a significant decrease in the level of RBCs, Hb, PCV and platelets.
B) Effect on total leukogram: Treatment with CYC alone once daily at a dose of (25 mg/kg b.wt.) for ten successive days provoked a significant decrease in the level of WBCs, neutrophil, lymphocyte and monocyte.
II- Effect of Vit C and/or Echinacea on immune response of cyclophosphamide treated mice.
A) Effect on phagocytic percent and phagocytic index. B) Effect on serum lysozyme activity. C) Effect on serum nitric oxide production. D) Effect on serum total protein, albumin and globulin. E) Effect on serum beta and gamma globulin levels.
III-Histopathological findings: Normal histological structure of the thymus, spleen and bone marrow from mice treated with Vit C and/or E. purpurea were noticed.
Spleen tissue sections of CYC treated group showed moderate depletion in the lymphoid contents of the white pulps. The affected structures suffered apoptotic and necrotic changes in the lymphocytes with increase in the intercellular spaces and in the fine eosinophilic reticular network and or deposition of fine albuminous materials (edema). The megakaryocytes were partially affected and showed necrotic or apoptotic changes in some cells. The splenic blood vessels were moderately congested. Examined sections of the spleen which obtained from the group co-treated with Vit C and CYC revealed mild depletion of the lymphoid elements of both white and red pulps due to apoptotic and necrotic changes in the affected cells. The red pulp showed activated enlarged histiocytes, may be a reactive processes following lymphoid depletion. The megakaryocytes appeared with a relative increase in their numbers. Examined section of the spleen which obtained from the group co-treated with E. purpurea and CYC showed moderate activity of the lymphoid cells of the white pulp as represented by increase number of mature and immature lymphocytes in the germinal centers and marginal zones.
