Abstract

Abstract
In the present study, A. fumigatus TXD105 (LC171332) and A. tenuissima TER995 (LC170562), two endophytic fungal strains, were positive for production of the anticancer compound, paclitaxel. In order to enhance paclitaxel production by the two strains, a series of culture conditions were examined under submerged fermentation. Moreover, Under optimized conditions, the final paclitaxel titer was intensified to 5.16- and 4.43-fold by the two respective strains when compared with their initial titers. The two fungal strains were then irradiated by UV, gamma and microwave rays and the third type showed less enhancement effect on paclitaxel production as compared with the first and second types of radiation. from the irradiation experiments, two stable mutants for paclitaxel production were obtained and designated as TXD105–GM6 and TER995–GM3. The two mutant strains were exploited in immobilization technique for paclitaxel production using different entrapping carriers. Several culture conditions were tried in immobilization to enhance paclitaxel production and the RSM optimization program was applied. As a result, the paclitaxel titer was intensified to 2.08- and 3.19-fold by the two respective strains, as compared by the initial titers of the free cultures.
