Abstract A total of 300 random samples including raw milk from mastitic cow (90), soft cheeses (Kareish, and Damietta ) 15 samples each, ice cream (30), and samples of human suffering from certain diseases S. aureus infection Samples were collected from different localities in Asyut City. The samples were examined bacteriologically for isolation, identification and molecular genotyping of S. aureus strain was done by using CoA gene S. aureus could be detected in 30-60% in the examined milk sample and milk product, and also it detected in 33-70 % of the human samples. Tube coagulase test was made for accurate identification of CoPS from CoNS.
