Abstract

Antimicrobial agents provide some of the most dramatic examples of the advances of modern medicine. This thesis is devoted to the development of new analytical methods for the determination of certain antibiotics; namely azithromycin dihydrate (AZT), roxithromycin (ROX), cefixime (CFX), cefdinir (CFD), nifuroxazide (NX), linezolid (LNZ) and levofloxacin hemihydrate (LEV). The thesis comprises six main parts:Part I: A general introduction about the studied drugs including: pharmacological actions, chemical structures, physical properties, and literature survey\. Part II: This part is divided into two parts: Part IIA: This part is devoted to develop simple, sensitive, specific and inexpensive spectrophotometric method for AZT and ROX in pharmaceutical preparations through metal complexation with Cu2+. Part IIB: This part is concerned with the development of a rapid, simple and sensitive spectrophotometric method for AZT and ROX in pharmaceutical preparations through reaction with N-bromosuccinimide. Part III: This part is divided into two parts: Part IIIA: This part represents a novel,sensitive, simple and validated spectrophotometric method for determination of CFX and CFD in their pharmaceuticals using Hantzsch reagent. Part IIIB: This part describes a sensitive, selective, economic, and validated spectrofluorimetric method for determination of CFX and CFD in their pharmaceuticals using Hantzsch reagent. Part IV: In this part, a new, simple and highly sensitive spectroflourimetric method for estimation of NX was proposed depending on reduction of the studied drug with Zinc powder in acidic medium. Part V: This part presents a reliable, accurate and sensitive micellar liquid chromatographic (MLC) method for simultaneous estimation of CFX in two of its combined pharmaceutical preparations with either LNZ or LEV. It was further extended to determine the concerned drugs in spiked human plasma without any extraction steps with high percentage recoveries. Part VI: This part aims to develop a rapid, simple, sensitive and isocratic MLC method using monolithic column for simultaneous estimation of NX and DV in their capsules. The developed method allowed the separation of the studied drugs with good resolution in a reasonable time.
