Abstract

Indomethacin is a well known non steroidal anti-inflammatory drug, it exerted many side effects including acute gastritis.
Montelukast is a selective leukotriene (LT) D4 receptor antagonist, anti-asthmatic and anti-inflammatory drug that interferes directly with LT production (5-lipoxygenase inhibitors).
The aim of the present study was to demonstrate the injurous effects of indomethacin on gastric mucosa of the stomach of adult albino male rat and to evaluate the possible protective role of montelukast in preventing the hazardous effects on the stomach.
Thirty-five adult male albino rats were fasted for 24 hours before drug ingestion to assure the full effect and dose of each drug. The rats were separated into five groups. Each group consisted of five rats.
Group(A) control: received normal saline through gastric gavage tube.
group (B): received indomethacin (48 mg/kg) through gastric gavage tube.
Group(C): received ranitidine(30 mg/kg) half an hour before receiving indomethacin (48 mg/kg) through gastric gavage tube.
group (D): received montelukast (5mg/kg) half an hour before receiving indomethacin (48 mg/kg) through gastric gavage tube.
Group(E): received montelukast (20mg/kg) half an hour before receiving indomethacin (48 mg/kg) through gastric gavage tube.
The rats were sacrificed 6 hours after drug ingestion, and their stomachs were dissected and opened along the greater curvature and examined by magnifying lens to calculate the ulcer index then it was examined by the light microscope.
The study revealed that indomethacin caused high ulcer index, extensive destruction of the mucosa,haemorrage,apoptotic bodies and inflammatory cells infiltration. Scanty amount of mucous secretion evidenced by negative PAS reaction.
In the group(C), receiving indomethacin and ranitidine, there was improvement of the morphology of the gastric mucosa in the form of: significant decrease of the ulcer index, almost normal gastric mucosal architecture. The amount of mucous secretion was increased evidenced by positive PAS reaction.
In the group(D), receiving indomethacin and montelukast(5mg/kg) , there was mild improvement of the morphology of the gastric mucosa in the form of, decrease of the ulcer index, and infrequent foci of superficial erosions associated with decreased inflammatory cells infiltration. The mucous secretion was increased evidenced by positive PAS reaction.
