Abstract

Cadmium chloride is heavy metals found in the environment, its classified as carcinogens, and humans exposed to its daily through many roads such as air, water, soil, Agriculture, Food, cigarette and tattoo.
The natural materials used in the prevention of the toxic effect of cadmium.
• Honey: an antioxidant and is used in the reduction of serious diseases such as cancer, immune deficiency diseases and natural medicine.
• Curcumin (Turmeric) which antioxidant and detoxifying where it is used in Chinese medicine.
The aim of this study
The aim of the present study using two natural products as an attempt to minimize the toxicity effect of cadmium.
In this study different methods were used to evaluate the cytogenetic effect of using cadmium.
Cytogenetic study
Detection of chromosome aberrations (structural, numerical and mitotic index).
Sperm head abnormalities
Determination of sperm abnormalities
Experiment design
Forty five (45) male Swiss albino mice were (MUS musculus) and weights ranging between 25-30 grams, thses mice were obtained from the National Research Center (N.R.C) in Dukki. Were divided into Nine groups, 5 mice each, as detailed below.
group (I), 5 mice Control group
group (II), 5 mice were injected intraperitoneally with 0.5ml saline solution daily for 96hs, as positive control.
group (III), 5 mice were injected intraperitoneally with 0.67mg CdCl2/ kg b. w. dissolved in 0.5 ml saline solution and give single dose for 96hs.
group (IV), 5 mice were injected intraperitoneally with 300mg HY /Kg b. w. dissolved in 0.5 ml saline solution and give daily for 96hs.
GroupV, 5 mice were injected intraperitoneally with 50 mg CMN /Kg b. w. dissolved in 0.5 ml saline solution and give daily for 96hs.
group VI, 5mice were injected intraperitoneally with honey daily for 96hs then single dose of CdCl2 for 96hs (as protective trial).
group IX, 5 mice were injected intraperitoneally with Curcumin daily for 96hs then single dose of CdCl2 for 96hs (as protective trial).
group X, 5 mice were injected intraperitoneally with single dose CdCl2 for 96hs then honey daily for 96hs (as treatment trial).
group XI, 5 mice were injected intraperitoneally with single dose CdCl2 for 96hs then curcumin daily for 96hs (as treatment trial).
Results
Structure aberration:
The highest percentage of chromosome aberrations observed in bone marrow cell of male mice was found clearly in the form of chromatid fragment, centric fusion, gaps and stickiness.
Numerical aberration:
Represented by the trisomy cells, there is highly significant increase with treated CdCl2.
Mitotic index:
The low rate of cell division in mice treated cadmium, cadmium before and after the curcumin protection and treatment and after honey as protection compared to the control, control positive, honey, curcumin and cadmium followed honey as treatment group.
Sperm abnormalities:
Four types of sperm head aberrations were recorded hummer, banana, amorphous and without hook the highest incidence of aberration in Cd group.
Conclusion
We conclude cadmium works to increase the rate of chromosomal abnormalities, reduce the rate of cell division in mice and decrease in account sperm with increase in number of sperm head abnormalities.
Honey and Curcumin have the same results control of chromosomes abnormalities & the rate of cell division while recorded increased in number normal sperm head.
Honey as protective or treatment and cadmium then curcumin as treatment group lead to decrease of chromosome aberration with increase in cell division that shows Cd HY, CdCMN and HY Cd could protect the body from cytotoxicity arising from cadmium and the same values of normal sperm were recorded.
While protection curcumin then cadmium have the same results of cadmium and it shows that curcumin could not protect the body from cytotoxicity arising from cadmium.
