Abstract

The object of the current thesis was for study the application of spray drying method, for produce microspheres triamcinolone acetonide (TAA) particles appropriate for inhalation and for study the impact of spray drying method on TAA. The spray drying method employed by the spray drying of the solution of TAA alone or with the blowing agent (for example ammonium bicarbonate , menthol and chloral hydrate) and TAA / polymer (for example PVP K-30 and HPβCD) from co-solvent system (90%(v/v) ethanol) for create the desired microspheres particles. Recovered particles were visualized by SEM and the physicochemical characteristics for the unprocessed and for spray dried specimens were had compared in terms at solid-state characterization; thermal behavior by DSC, p-XRD and FTIR. Micromeritics characteristics for all samples were investigated. These included evaluation of particle size, bulk and tapped densities and carrś index. In-vivo deposition spray dried powders of TAA alone were evaluated. The morphology of recovered particles was dependent on spray drying parameters/conditions, which included changes in feed concentration, changes in solvent concentration, changes in type and concentration of the blowing agent and changes in type and concentration of polymer employed. Spray dried TAA with or without the blowing agent demonstrated superior aerodynamic properties relative to micronized TAA. The solid state of TAA was not alter on spray drying with or without the blowing agent but the solid state of TAA was alter on spray drying with polymer. The solid state was produced regardless of the blowing agent type and concentration used. This was not different from previous studies which showed that blowing agents were evaporated upon spray drying. Microspheres of triamcinolone acetonide with or without the blowing agent and with polymer were produced on spray drying from 90 % (v/v) ethanol system. Particle characterization of recovered microspheres particles showed favorable characteristics for pulmonary delivery compared to micronize triamcinolone acetonide. TAA were remained with curative concentrations in the tissue of lung rats for at the least 24h next insufflation. This suggests potential for future work and development of these systems, as a therapeutically used TAA suitable for pulmonary delivery.
