Abstract

The presented thesis was concerned with analytical studies of some cephalosporins namely: (CFX), (CPD), (CFZ) and (CPM). The presented thesis falls in three parts:
Part I: General Introduction
This part provides a general introduction about the investigated compounds such a pharmaceutical importance, chemical structure and analytical review for the analytical techniques used for determination of the studied drugs in pure forms and dosage forms, at the end of this part we described the objective of the suggested work.
Part II: was divided into 3 chapters.
Chapter 1: The preparation and characterization of the metal complexes of CFX, CPD and CFZ with Al (III), Ca (II), Sr (II) and Zn (II) were presented for the first time. The existence of these complexes has been confirmed by both UV/Vis and FTIR spectroscopy. According to the FTIR spectroscopic data, different mechanisms were suggested for the coordination of the studied drugs with these metal ions. The in vitro evaluation of these complexes has revealed a significant reduction of their antimicrobial activities for almost all the studied metal complexes; therefore, we recommend that these metals should not be administered with the studied cephalosporins. Furthermore, the formation metal complexes have been utilized for the development of quantitative atomic absorption spectroscopic method which could be applied for the determination of cited drugs in their commercially dosage forms.
Chapter 2: The key FTIR spectral features of each of the drug investigated and their combinations were reliably determined. The FTIR spectroscopy has been utilized for the first time to quantify the studied drugs and their binary mixtures by integrating the peak areas of their diagnostic and well-resolved bands. The results were compared with other relevant spectrophotometric methods, the obtained results have provided good sensitivity, good recovery and linearity range. This vibrational spectroscopic technique appears to be an efficient, simple and rapid tool to quantify the active ingredients present in a complex matrix in the pharmaceutical formulations.
Chapter 3: A novel facile reductant method to produce antibiotic-coated gold and silver nanoparticles for colorimetric determination of CPM and CFZ was described. Interestingly, the conjugations of CPM and CFZ with silver and gold nanoparticles have significantly enhanced their antibacterial activity.
