Abstract

The present work was performed to study the protective and curative anti-inflammatory role of natural grape seed extract (GSE) and indomethacin as standard anti-inflammatory drug alone or/ in combination on Carrageenan-induced paw edema in rats by measurement of selective pro-inflammatory cytokines. Moreover the side effects of these substances were studied.
One hundred and ten adult male Sprague-Dawley rats were divided equally into 11 groups. group (1) was Kept as normal control. group (2) was received GSE (40 mg/kg b.wt) orally for 10 days. group (3) was received indomethacin (5 mg/Kg b.wt) orally for 10 days. group (4) was received both indomethacin (5 mg/Kg b.wt) and GSE (40 mg/kg b.wt) orally for 10 days. Group
(5) was received a single injection of 0.1 ml of carrageenan-saline solution (1%, w/v) into the plantar surface of the hind paw of left leg, at the 10th day of the experiment. Rats of groups (6,7 and 8) given GSE, indomethacin and their combination respectively with the previous doses then after last dose by 2 hours, paw edema was induced. Meanwhile rats in groups (9,10 and11) were injected with carrageenan for paw edema induction then after one hour treated with a single dose of GSE, indomethacin and their combination respectively with the previous doses. Edema volume was recorded after1, 2, 3, 4 and 5 hours from carrageenan injection, and then animals were killed for blood and tissue sampling to perform the hematological, biochemical and histopathological studies.
