Abstract

The objectives of this study were to determine the negative impact of two commonly used food preservatives (BHA and Sodium Benzoate). FDA (Food and Drug Administration) of USA release annually a list of banned food additives. However, the clinical, cytological, genetic proofs are still under continuous evaluation to decrease the seriousness of the prevention strategies. In developing countries, lack of scientific evidence and coordination influence the passive acts of industrial quality controllers and observers. Systems biology approaches (Knock-out Yeast) and human colon cancer cell line were effective tools in determining the effects of these two food additives on the expression of some human cancer-related genes homologous to selected yeast strains (genotypes)
The obtained results could be summarized as follows:
1- Selected Knock-out Yeast’s strains were treated with different concentrations of each of the two food additives under study for 24 hr. and their growth curve were estimated. Growth curve of selected yeast strains (genotypes) showed negative impact on growth rates, In additives, YKO strains confirmed the occurrence of cancer formation due to administration of these food additives that could be caused according to different genotypes. This might give an overview of the relationship between those additives and cancer formation.
2- Colon Cancer Cell Line were treated with the same concentrations of food additives under study for 24 hr. Viability in shapes of the cells showed considerable variations between control and treatment by NRU assay and effect of food additives on them and confirmed the carcinogenic effect of these additives.
3- Real-Time PCR of cancer-related genes of cell line homologous with selected genotypes were done. Results of gene expression of selected genotypes were on human genes showed that the effect of additives could inflict deletarious and pronounced health hazards in human including cancer.
4- Prediction of Protein-Protein interaction was done by String web site. Results of yeast protein interaction showed some links between selected genes on yeast (10 genes) in two interaction links, first link between four genes which were related with signals of cell cycle and other link between two genes were related with MTAP family (that control tumor formation). Results of human protein-protein interaction showed the same homologous genes with those of yeast which confirmed the relationship between the two systems.
5- In conclusion, it is evident that the food industry should adhere to the rules and recommendation set by FDA and other health organizations with regard to the optimum concentrations of such food additives to avoid any hazards health impacts on human.
