Abstract

This study aimed to investigate the protective effect of olive oil on atherosclerot diabetic rats. This study is on thirty male albino rats divided into three equal groups ten each group. The first group fed normal control diet, it was the control –ve group , the second and the thired groups were type II diabetis induced by high fat diet for 2 weeks followed by a single low dose intraperitoneal injection of streptozotocin (STZ) (35mg/kg, dissolved in 0.05M citrate buffer,pH4.5) then they received atherosclerotic diet for 30 consecutive days with oral administration of olive oil to the third group only (at dose 0.5 ml for each rat) daily from the start of the experiment. At the experiment end period , blood samples collected by heart puncture and used for determination of blood glucose level , serum insulin, serum lipid profile (serum total cholesterol, serum triacylglycerol, serum HDL-C and serum LDL-C) , lipid peroxidation (MDA) , antioxidant activity of glutathione peroxidase (GPx) and RT-PCR of leptin gene expression .The obtained results showed that olive oil supplementation resulted in a significant decrease in serum total cholesterol, triacylglycerol and LDL-c with a significant increase in HDL-c in addition to significantly decreasing the serum level of lipid peroxide (MDA), also olive oil supplementation found to significantly increase the level of both serum glutathione peroxidase activity and serum insulin level with decreasing trend in the glucose level as a result of olive oil supplementation. As well as there was a downregulation in the leptin gene expression. from this study we concluded that olive oil has a protective effect on the atherosclerosis development in diabetic induced rats.
