Abstract The aim of this study was screening the highest production of protease by bacteria isolated from a poultry waste soil. The isolated bacterial protease was partially purified and its kinetic properties were studied. Eight selected bacterial isolates were identified by both Gram stain and biochemical Test via strip system Vitek 2C. Staphylococcus sciuri, which showed the highest protease activity was selected for enzyme production and purification. The enzyme was purified using ammonium sulphate precipitations and sephacryl S200 gel filtration chromatography column. The molecular weight of S.sciuri protease was determined by using SDS-PAGE electrophoresis. Subsequent characterization of the enzyme fraction with the highest activity was carried out. The obtained results indicated that the enzyme obtained by 80 % ammonium sulphate fractionation exhibited a purification fold and recovery yield of 2.23 and 18%, respectively. Three isozymes (I, II and III) were determined using Sephacryl S200 column, the highest activity was exhibited by isozyme I with 27.5 purification fold, 4% yield and a molecular weight of 15KDa. |
