Abstract Formulation and Evaluation of Tablets Containing the Hypolipidemic Drug Atorvastatin Calcium Atorvastatin calcium (ATV) is a hyopolipidemic drug used in the primary and secondary prevention of cardiovascular diseases in patients with high levels of cholesterol. It acts by inhibiting HMG-CoA reductase hence blocking the pathway for synthesizing cholesterol in the liver. ATV is practically insoluble in water and belongs to Class II drugs according to the biopharmaceutical classification system having poor solubility and high permeability. The bioavailability of ATV is as low as 14% when administered orally due to its limited dissolution and because it is subjected to first pass metabolism in the gut and liver. In the present work preformulation studies were carried out using different excipients and techniques to detect any incompatibility with the used excipients and to choose the most effective systems of enhancing its dissolution with acceptable drug and formulation stability. The preformulation study used differential scanning calorimetry (DSC), differential thermal analysis (DTA), thermogravimetric analysis (TGA). Fourier-transform Infrared (FT-IR) and X-ray diffraction (XRD). |
